Peptide Purity vs Identity: Why a Percentage Is Not Enough

The difference between chromatographic purity and confirmed identity, why a single percent can mislead, and how a research record should combine both.

Direct answer

Purity and identity are different properties. Purity usually describes how much of a sample appears as a single chromatographic peak, while identity confirms that the peak is the intended sequence. A high purity percentage only means the chromatogram is clean; it does not prove the material is the named peptide or that it is fit for a research workflow. A complete batch record reports both, plus lot traceability.

Two questions that are often confused

'How pure is this vial?' and 'Is this vial the peptide I ordered?' are separate questions. Purity is usually a chromatographic measurement from HPLC. Identity is usually a mass measurement from mass spectrometry or a sequence-specific method.

A material can be highly pure (one dominant peak) and still be the wrong sequence, a related byproduct, or a degraded form. Conversely, a correct identity result does not tell you how clean the bulk sample is.

Why a single number is not a quality record

  • A purity percentage does not name the compound
  • A purity percentage says nothing about net peptide content
  • Identity by mass does not establish chromatographic purity
  • Neither result addresses sterility, endotoxin, or assay suitability
  • Without a lot identifier the result cannot be tied to the vial in hand

How to read both together

A stronger research record pairs an HPLC purity result with an MS identity result and a lot identifier, all tied to the certificate for that batch. Researchers can then compare the vial, the lot, and the report without guessing, and retain the lot information with their own records.

If only a purity number is published, treat it as one signal among several. Ask the supplier for the identity method and the lot before relying on the material for published or replicated work.

What this means in practice

Researchers evaluating research peptides should ask for both pieces of documentation for every lot, not a single headline number. A supplier that publishes per-batch CoAs has already done part of this work for the laboratory.
