Net Peptide Content Explained: Weight, Purity, and What's in the Vial

Net peptide content is the fraction of the vial's total weight that is actual peptide, as opposed to water, counterions, salts, or residual solvents. It is a separate measurement from HPLC purity, which only describes the peptide fraction of the chromatogram. Two samples can have the same purity percentage and very different net peptide content, so researchers should review both the content value and the purity value for a lot.
Why the label weight is not the peptide weight
A vial labeled with a nominal weight typically describes the gross content of the vial, not the amount of active peptide. After lyophilization, peptides retain variable amounts of water and counterions, and some material is salts or residual solvent rather than peptide.
Net peptide content is determined separately, often by elemental analysis or amino-acid analysis, and expresses how much of the labeled weight is actually the peptide of interest.
Purity and content answer different questions
- Purity: of the peptide material in the chromatogram, how much is the target peak
- Content: of the total weight in the vial, how much is peptide at all
- High purity with low content means a clean sample but little active peptide
- High content with low purity means most of the weight is peptide that is not the target
- A research workflow needs both numbers to prepare a known amount of the intended sequence
What to look for in a certificate
A complete record reports the lot identifier, the HPLC purity method, the net peptide content method, and a clear statement of how the values were obtained. Where a supplier publishes both, a laboratory can convert label weight into an approximate amount of target peptide for its own workflows.
If only a purity percentage is published and no content value is given, treat any mass-based preparation as approximate and confirm the content with the supplier before relying on it in published work.
